First, one needs OMS (Opcode Music System), which I believe is free:

The author of the MPX-100 Editor, Jon Witte, says this:
Any interface that works with OMS will work, which most of them do. It doesn't matter if it's serial or USB as long as it's OMS compatible. The MPX-100 Editor requires OMS to work. Check out these pages for some midi
