Assuming your input drivers and output are selected properly, my first guess would be that you have a 64 bit operating system. roland sound canvas dxi doesn't work with 64 bit.
If it's a 32 but system then check drivers.

If you want to run an internal soft synth, you can use the coyote forte system from PG Music. Or if you want to go external you can use something like a Roland SD20. I use the forte most often and it works well.
